    Are Southern Resident killer whales on a path to extinction?

    Numerous extinctions reveal species tipping points where multiple pressures converge to cause a cascade to demise. This paper examines how multiple pressures impacting Southern Resident Killer Whales (SRKW) could lead to their rapid downward spiral to extinction. The most commonly cited impacts to this population are historically low abundance of chinook salmon prey, vessel disturbance from ships and boats, toxicant loads above established health effects thresholds, and risk of oil spills from increased vessel and rail oil transport. Inbreeding and a heavily skewed male biased sex ratio at birth compound these problems. Lack of prey increases nutritional stress and dependence on fat reserves, while decreasing concentrations of bioactive thyroid hormone (T3). Fat metabolism increases circulating lipophilic toxins, which can further decrease T3 concentrations. Collectively, these impacts appear to increase rates of spontaneous abortion and neonatal loss. They may also lead to immunosuppression with a likely rise in incidence of disease and mortality. Associated population decline inherently increases inbreeding, which can increase male sex ratio bias at birth. Reduced T3 may also affect sex ratio at birth. SRKW seasonal presence in the Salish Sea is declining, attributed to declining prey and increased whale-watch vessels. Breeding events within and between pods are commonly observed during these visits to the inland waters. Their decline in visits may limit opportunities for breeding with a more diverse gene-pool, which could further increase inbreeding and associated potential for male-biased sex ratio bias at birth. Chinook remain at the epicenter of all this, indicating an increase in Chinook salmon abundance to be the single most effective mitigation we can do to recover SRKWs. Salmon are sentinels of ecosystem imbalance spanning from toxic contaminants to habitat destruction. Creative and immediate solutions are essential to expedite the process of recovery and ensure the survival of this iconic species

    Performance test of QU-fitting in cosmic magnetism study

    QU-fitting is a standard model-fitting method to reconstruct distribution of magnetic fields and polarized intensity along a line of sight (LOS) from an observed polarization spectrum. In this paper, we examine the performance of QU-fitting by simulating observations of two polarized sources located along the same LOS, varying the widths of the sources and the gap between them in Faraday depth space, systematically. Markov Chain Monte Carlo (MCMC) approach is used to obtain the best-fit parameters for a fitting model, and Akaike and Bayesian Information Criteria (AIC and BIC, respectively) are adopted to select the best model from four fitting models. We find that the combination of MCMC and AIC/BIC works fairly well in model selection and estimation of model parameters in the cases where two sources have relatively small widths and a larger gap in Faraday depth space. On the other hand, when two sources have large width in Faraday depth space, MCMC chain tends to be trapped in a local maximum so that AIC/BIC cannot select a correct model. We discuss the causes and the tendency of the failure of QU-fitting and suggest a way to improve it.Comment: 8 pages, 9 figures, submitted to MNRA

    Automated image processing is a critical and often rate-limiting step in high-content screening (HCS) workflows. The authors describe an open-source imaging-statistical framework with emphasis on segmentation to identify novel selective pharmacological inducers of autophagy. They screened a human alveolar cancer cell line and evaluated images by both local adaptive and global segmentation. At an individual cell level, region-growing segmentation was compared with histogram-derived segmentation. The histogram approach allowed segmentation of a sporadic-pattern foreground and hence the attainment of pixel-level precision. Single-cell phenotypic features were measured and reduced after assessing assay quality control. Hit compounds selected by machine learning corresponded well to the subjective threshold-based hits determined by expert analysis. Histogram-derived segmentation displayed robustness against image noise, a factor adversely affecting region growing segmentation

    Influence of restraining devices on patterns of pediatric facial trauma in motor vehicle collisions.

    In the Commonwealth of Pennsylvania, it is required that all children under the age of 4 years be restrained by an infant seat or car seat appropriate for their age and weight. Furthermore, all individuals riding in the front seat must be restrained by a seatbelt. This study examined the relationship between patterns of facial injuries and the use of restraining devices in the pediatric population. A retrospective analysis was performed on motor vehicle collision data submitted to the Pennsylvania Trauma Outcome Study database from 1990 through 1995. Criteria for submission included trauma patients who were admitted to the Intensive Care Unit, those who died during hospitalization, those who were hospitalized for more than 72 hours, or those who were transferred in or out of the receiving hospital. A subset of 412 pediatric patients, 15 years of age or younger, was analyzed for patterns of facial injury and the presence or absence of restraining devices. Restraining devices were categorized as a car seat or a seatbelt. Statistical analysis was performed using chi-square and Fisher\u27s exact tests. Of the 412 pediatric patients, only 17 children were restrained with a car seat and 121 were wearing a seatbelt. A total of 30 children sustained facial fractures, and 50 children suffered facial lacerations. There was a statistically significant increase in the incidence of facial fractures with increasing age of the child (p \u3c 0.001). Of children with facial fractures, 70 percent of those 5 to 12 years old and 90 percent of those 13 to 15 years old were unrestrained (p = 0.166). In conclusion, despite legislation mandating the use of restraints, a large proportion of children involved in motor vehicle collisions were unrestrained. Furthermore, there seems to be a direct relationship between the age of a child and the incidence of facial fractures sustained in motor vehicle collisions

    Comparison of growth rates of the filamentous fungus Aspergillus niger L. from contrasting environments

    Eighteen strains of filamentous fungus Aspergillus niger were isolated from contrasting environments in Israel: the hypersaline Dead Sea and the temperate «Evolution Canyon» I. A comparison of growth rates under different water activity stress was provided. Clear differences in growth rates were observed under 5—20 % and 40 % volumes of the Dead Sea water (DSW). No difference was observed when we used 25— 35 % volumes of DSW. Strains from «Evolution Canyon» I grown at lower concentrations of the DSW revealed higher growth rates, while strains from the DSW demonstrated higher activity under higher stress (40 % volumes of DSW).     The Extrachromosomal EAST Protein of Drosophila Can Associate with Polytene Chromosomes and Regulate Gene Expression

    The EAST protein of Drosophila is a component of an expandable extrachromosomal domain of the nucleus. To better understand its function, we studied the dynamics and localization of GFP-tagged EAST. In live larval salivary glands, EAST-GFP is highly mobile and localizes to the extrachromosomal nucleoplasm. When these cells are permeabilized, EAST-GFP rapidly associated with polytene chromosomes. The affinity to chromatin increases and mobility decreases with decreasing salt concentration. Deleting the C-terminal residues 1535 to 2301 of EAST strongly reduces the affinity to polytene chromosomes. The bulk of EAST-GFP co-localizes with heterochromatin and is absent from transcriptionally active chromosomal regions. The predominantly chromosomal localization of EAST-GFP can be detected in non-detergent treated salivary glands of pupae as they undergo apoptosis, however not in earlier stages of development. Consistent with this chromosomal pattern of localization, genetic evidence indicates a role for EAST in the repression of gene expression, since a lethal east mutation is allelic to the viable mutation suppressor of white-spotted. We propose that EAST acts as an ion sensor that modulates gene expression in response to changing intracellular ion concentrations

    The biogeography of the caribou lungworm, Varestrongylus eleguneniensis (Nematoda: Protostrongylidae) across northern North America

    Varestrongylus eleguneniensis (Nematoda; Protostrongylidae) is a recently described species of lungworm that infects caribou (Rangifer tarandus), muskoxen (Ovibos moschatus) and moose (Alces americanus) across northern North America. Herein we explore the geographic distribution of V. eleguneniensis through geographically extensive sampling and discuss the biogeography of this multi-host parasite. We analyzed fecal samples of three caribou subspecies (n = 1485), two muskox subspecies (n = 159), and two moose subspecies (n = 264) from across northern North America. Protostrongylid dorsal-spined larvae (DSL) were found in 23.8%, 73.6%, and 4.2% of these ungulates, respectively. A portion of recovered DSL were identified by genetic analyses of the ITS-2 region of the nuclear rDNA or the cytochrome oxidase c subunit I (COI) region of the mtDNA. We found V. eleguneniensis widely distributed among caribou and muskox populations across most of their geographic prange in North America but it was rare in moose. Parelaphostrongylus andersoni was present in caribou and moose and we provide new geographic records for this species. This study provides a substantial expansion of the knowledge defining the current distribution and biogeography of protostrongylid nematodes in northern ungulates. Insights about the host and geographic range of V. eleguneniensis can serve as a geographically extensive baseline for monitoring current distribution and in anticipating future biogeographic scenarios under a regime of accelerating climate and anthropogenic perturbation

    Distinguishing the Impacts of Inadequate Prey and Vessel Traffic on an Endangered Killer Whale (Orcinus orca) Population

    Managing endangered species often involves evaluating the relative impacts of multiple anthropogenic and ecological pressures. This challenge is particularly formidable for cetaceans, which spend the majority of their time underwater. Noninvasive physiological approaches can be especially informative in this regard. We used a combination of fecal thyroid (T3) and glucocorticoid (GC) hormone measures to assess two threats influencing the endangered southern resident killer whales (SRKW; Orcinus orca) that frequent the inland waters of British Columbia, Canada and Washington, U.S.A. Glucocorticoids increase in response to nutritional and psychological stress, whereas thyroid hormone declines in response to nutritional stress but is unaffected by psychological stress. The inadequate prey hypothesis argues that the killer whales have become prey limited due to reductions of their dominant prey, Chinook salmon (Oncorhynchus tshawytscha). The vessel impact hypothesis argues that high numbers of vessels in close proximity to the whales cause disturbance via psychological stress and/or impaired foraging ability. The GC and T3 measures supported the inadequate prey hypothesis. In particular, GC concentrations were negatively correlated with short-term changes in prey availability. Whereas, T3 concentrations varied by date and year in a manner that corresponded with more long-term prey availability. Physiological correlations with prey overshadowed any impacts of vessels since GCs were lowest during the peak in vessel abundance, which also coincided with the peak in salmon availability. Our results suggest that identification and recovery of strategic salmon populations in the SRKW diet are important to effectively promote SRKW recovery

    We consider procurement of an innovation from heterogeneous sellers. Innovations are random but depend on unobservable effort and private information. We compare two procurement mechanisms where potential sellers first bid in an auction for admission to an innovation contest. After the contest, an innovation is procured employing either a fixed prize or a first-price auction. We characterize Bayesian Nash equilibria such that both mechanisms are payoff-equivalent and induce the same efforts and innovations. In these equilibria, signaling in the entry auction does not occur since contestants play a simple strategy that does not depend on rivals' private information
